Melanotan 1 (afamelanotide) is a synthetic analog of the naturally occurring α-melanocyte-stimulating hormone (α-MSH). It stimulates melanin production to promote skin tanning and is prescribed under the brand name Scenesse primarily to protect against UV-induced skin damage in patients with rare light sensitivities like erythropoietic protoporphyria (EPP).
- Mechanism: It binds to the melanocortin-1 receptor (MC1R), which increases the production of eumelanin (the pigment that darkens skin).
- Photoprotection: By increasing melanin, it provides an internal barrier against ultraviolet radiation, significantly reducing sunburn and cellular DNA damage.
- Medical Application: It is approved by regulatory bodies (such as the FDA in the US and EMA in Europe) as an implantable, controlled-release formulation (Scenesse) for EPP.
Melanotan 1 vs. Melanotan 2
- Melanotan 1 (Afamelanotide): Highly selective to MC1R. Primarily used to induce tanning for photoprotection with fewer systemic side effects.
- Melanotan 2: A shorter, non-selective peptide. It often induces tanning at lower cumulative doses but is known for causing more adverse side effects, including severe nausea, spontaneous penile erections, and changes in moles (which can increase melanoma risk)
